Semaglutide reconstitution calculator.
The FDA-approved semaglutide products, Ozempic and Wegovy, ship as pen injectors that are already in solution, so there is nothing to reconstitute. Lyophilized semaglutide vials come from compounding and research supply, and that is the only context where this arithmetic applies.
Reference range often cited for Semaglutide: 250–2400 mcg, shown for orientation only, not a recommendation. Enter your own target below. Read the evidence →
Each unit on a 100u · 1.0 mL syringe ≈ 15 mcg of this solution.
Show the math
More water → lower concentration → more units for the same amount.
What changes the math for Semaglutide.
- Vials are commonly 3 mg, 5 mg, and 10 mg. A 5 mg vial in 2 mL is 2,500 mcg/mL, so 250 mcg is 10 units and 1 mg is 40 units.
- Labeled semaglutide amounts are written in mg per week (0.25 mg, 0.5 mg, 1 mg, and up), which is milligram-scale arithmetic on a vial measured the same way. Entering mg directly avoids a conversion error.
- Compounded copies of an approved drug are only permitted in specific regulatory circumstances, and that status has changed over time. The legal status page covers where semaglutide actually stands.
- The approved titration schedule is a published regulatory fact and is reproduced on the GLP-1 titration calculator. It is not a recommendation from this site.

Weekly SC injection. Wegovy titration is 5 steps over 16+ weeks.
FDA-approved semaglutide is available as Ozempic (diabetes, weekly) and Wegovy (weight, weekly), both as pen injectors for subcutaneous injection. The Wegovy titration schedule below is reproduced from the FDA prescribing information, a public regulatory fact.
Ozempic (diabetes indication) uses 0.5 mg, 1 mg, and 2 mg doses: a different schedule and dose ceiling from Wegovy. Do not interchange the two products.
If a dose step is not tolerated (persistent nausea/vomiting), the prescribing information allows delaying the next escalation. Your prescriber decides timing.
FAQ.
Do I need to reconstitute Ozempic or Wegovy?
No. Both are supplied as pen injectors containing solution, ready to use as labeled. Reconstitution only applies to lyophilized powder, which is how compounded and research-supply semaglutide is presented.
How many units is 0.25 mg of semaglutide?
On a 5 mg vial in 2 mL (2,500 mcg/mL), 0.25 mg is 250 mcg, which is 10 units on a U-100 syringe. Change either the vial or the water and that number changes, so enter your own.
Is a U-100 insulin syringe right for semaglutide?
U-100 marks volume, not insulin content: one unit is 0.01 mL regardless of what is in the barrel. The reading is only meaningful once you know your own mcg per mL, which is what the calculator establishes.
This page is arithmetic on numbers you enter. It does not recommend an amount of Semaglutide, and nothing here is medical advice. For what the published evidence actually supports, read the Semaglutide profile, and for regulatory status see is Semaglutide legal. Reviewed June 1, 2026.
Different question? The general reconstitution calculator covers every compound on the site, the blend calculator handles multi-peptide stacks, and vial longevity turns a vial into weeks of supply.
